Markle will be interviewed first, husband Harry will join later
Winfrey is a close friend of Markle and had attended her glittering wedding ceremony in 2018.
As per the format, the host will first speak with the Duchess of Sussex and the Duke will join later.
The special interview is being produced by Harpo Productions. Terry Wood and Tara Montgomery are working as executive producers on it while Brian Piotrowicz is a co-executive producer.

The US broadcaster said, "Winfrey will speak with Meghan, the Duchess of Sussex, in a wide-ranging interview, covering everything from stepping into life as a royal, marriage, motherhood, philanthropic work to how she is handling life under intense public pressure."
Once Harry joins, the couple will talk about their decision to shift to the US, their "future hopes and dreams for their expanding family."
Buckingham Palace has no sway over the couple's moves now
It is not known if the couple informed Buckingham Palace about this rare and much-anticipated interview. Since they are non-working members of the monarchy, the couple doesn't need to seek permission for their press obligations.
The palace has declined to comment on this development.
Meanwhile, the Queen is expected to review Harry's honorary military titles before the first anniversary of their unprecedented decision.
Not long ago, the couple announced Markle's second pregnancy
The announcement comes on the back of another big news linked to the Sussexes. On Sunday, they announced Markle was pregnant with her second child.
"We can confirm that Archie is going to be a big brother. The Duke and Duchess of Sussex are overjoyed to be expecting their second child," a statement read.
Markle had gone through an unfortunate miscarriage in July 2020.
