Sankranthiki Vasthunam narrates the story of YD Raju (Daggubati), who is assigned the job of finding Satya Akella, a kidnapped CEO of a multinational company. Things get complicated when Raju's wife Bhagyalakshmi insists on accompanying him on the rescue mission as she suspects him of having an affair with his ex-girlfriend ACP Meenakshi IPS. Besides Daggubati, Meenakshi Chaudhary and Aishwarya Rajesh also play key roles in the film.
